What Is Glutathione?
Glutathione (GSH) is a tripeptide of glutamate, cysteine, and glycine, distinguished by an unusual gamma-peptide bond between glutamate and cysteine that makes it resistant to ordinary peptidase breakdown. It is the most abundant intracellular antioxidant in the body, present in millimolar concentrations in most cells, and functions as the cell's primary redox buffer — the reserve that keeps the intracellular environment in a reduced, protected state.
Mechanism of Action
Glutathione works through a reduced (GSH) / oxidized (GSSG) cycle. The ratio of the two is one of the most-used biochemical markers of oxidative stress in research:

- Redox signaling and protein protection. Reversible glutathionylation of protein thiols regulates enzyme activity and shields cysteine residues from irreversible oxidative damage.
Research Evidence — and the Delivery Debate
The central research question with glutathione is not whether it matters — it plainly does — but whether supplementing it actually raises tissue levels. Orally administered glutathione is substantially broken down in the gut, and early studies questioned whether it meaningfully raised body stores at all.
A frequently-cited randomized controlled trial (Richie et al., 2015) found that sustained daily oral glutathione did raise body stores of glutathione in blood and tissues over 6 months, partially countering the older assumption that oral dosing is futile — though effect sizes and the best route remain actively debated. Liposomal, sublingual, intravenous, and precursor-based strategies (e.g., N-acetylcysteine to supply cysteine, the rate-limiting substrate) all exist precisely because the delivery question is unresolved.
Beyond redox biology, glutathione has been studied in hepatic conditions, Parkinson's disease (small IV trials), and dermatology, where it inhibits tyrosinase and has been marketed for skin lightening — an application with weak, contested evidence and real safety concerns around unregulated injectables.
